چکیده

DNA double helices comprising two neighbouring artificial imidazole-Ag(I)-imidazole base pairs incorporate the Ag(I) ions in a cooperative fashion. Isothermal titration calorimetry data confirm the theoretically predicted binding stoichiometry and indicate that Ag(I) binds with association constants >10(6) M(-1) to the metal-free DNA duplex.

Formation of Silver Nanoclusters from a DNA Template Containing Ag(I)-Mediated Base Pairs.

A series of DNA double helices containing different numbers of silver(I)-mediated base pairs involving the artificial nucleobases imidazole or 2-methylimidazole has been applied for the generation of DNA-templated silver nanoclusters. The original Ag(I)-containing nucleic acids as well as the resulting nanoclusters and nanoparticles have been characterized by means of UV/Vis spectroscopy, circu...

Solution structural studies of the Ag(I)-DNA complex.

We report equilibrium dialysis and electric dichroism studies of the two strong complexes (I and II) of silver ion with DNA. Cooperative conversion of DNA to the stronger type I complex results in a 9% length decrease, and a structure in which intercalated ethidium is perpendicular to the helix axis. Upon addition of more Ag+ to form the type II complex, the DNA length reverts to its original v...

Ag+-mediated DNA base pairing: extraordinarily stable pyrrolo-dC-pyrrolo-dC pairs binding two silver ions.

6-Substituted pyrrolo-dC-pyrrolo-dC mismatches selectively capture silver ions to form extraordinarily stable metal-mediated base pairs. One single modification in a 12-mer duplex causes a Tm increase of 36.0 °C relative to the metal-free mismatched duplex. Spectrophotometric titrations as well as ESI mass spectra confirmed the binding of two silver ions per base pair. The Ag(+)-mediated base p...

Silver(I)-mediated Hoogsteen-type base pairs.

Metal-mediated Hoogsteen-type base pairs are useful for the construction of DNA duplexes containing contiguous stretches of metal ions along the helical axis. To fine-tune the stability of such base pairs and the selectivity toward different metal ions, the availability of a selection of artificial nucleobases is highly desirable. In this study, we follow a theoretical approach utilizing disper...

Unprecedented dinuclear silver(I)-mediated base pair involving the DNA lesion 1,N(6)-ethenoadenine.

The DNA lesion 1,N(6)-ethenoadenine (εA) has been investigated with respect to its metal-binding properties. A synthetic DNA duplex comprising an εA : εA mispair readily forms doubly silver(I)-mediated base pairs εA-Ag(I)2-εA, representing the first example for a dinuclear metal-mediated homo base pair of a purine derivative. It also constitutes the first example for a Hoogsteen-type metal-medi...
